Like most portrays as 2P Japan that you'd see in 2P Hetalia, Dark Japan is Kiku's 'dark side' or 'evil twin' that makes Kiku or does the bad stuff that Japan (the country) has done throughout his history.

Dark Hetalia is where the character is insane, effected by war or during the war, it's usually bloody, gorey or tragic.
2p! is alternative colours that Himu made, the fandom decide to give them different personalities. In other words they're from a different universe and that's all.
So no, do your research.

I hate 2p!Japan because it's not even 2p!. It's normal Japan which the fandom decided to dub 2p. The Japanese wore a black uniform more often than the white. B/c of the fandom now everyone thinks it's his 2p. ugh.

2p!: red eyes
