The reason Loki cares is because the Tessact is influencing him to do these evils things, and a part of him doesn't WANT to! And when does do 'em, it wasn't thrilling.

That, and the fact that the Mind Gem's in his possession.  The Mind Gem controls the minds of both its wielder(s) (Loki) and victims (Hawkeye and the doctor guy).  Thanos knew about the Mind Gem's abilities, and he thought Loki was the perfect candidate for it.

Don't fret, I didn't figure it out either, until much later when I watched Guardians of the Galaxy (coughalsolookedituponlinecough).
They mention the Infinity Gems in GotG, it's the scene when they meet the Collector.
