If you mean the transparency, simply paint it plain white at first, and then erase it slowly with soft eraser.
For the tools, I usually use Redjuice's brush for characters and JonasDeRo's brushes for backgrounds.

I usually just use my feeling, lol.
Well, the brush I use the most are CSP's Mapping Pen. I use Redjuice's watercolor brush if I wanted smoother result. For backgrounds, mostly Jonas de Ro's cloud brushes, vegetation brushes, and water brushes.
