That's the end of the penultimate scene of this episode. Only the final one to go!
The "Kara Motif" is heard during the teaser to TOS "Spock's Brain" when Kara materializes on the bridge, and then is used for her as a leit motif (aka personal theme) throughout the episode. It's only nine seconds long, not counting the silence bumpers, but it's unmistakable. It's a dreamy little motif, just as the dreamy thing that's about to happen to Uhura here, when she gets kissed by Kirk as part of his way of saying thanks for the surprise party instead of being spanked. Here's a YouTube link to the scene in question from the episode; you can hear the "Kara Motif" starting at 01:03 in the clip and ending at 01:22:

"How's It Going?" is a short alternate take on Gem's theme from TOS "The Empath" that you hear about two-thirds the way through the episode. I'm using it here to highlight the deep bond of friendship (not romance!) that has developed between Kirk and Uhura during their time together on the Enterprise, and in particular following the events of TOS "Plato's Stepchildren" about a year or so prior to this tale. This particular BGM from TOS "The Empath" is not available online at this time and can only be found as part of the very pricey 15-disc TOS Soundtrack Complete Collection. The first 28 seconds of "Time Grows Short" from the common Fred Steiner/Royal Philharmonic re-recording by Vanese Sarabande records is an acceptable substitute, as that recording contains most of the episode's music and it's also another variation on Gem's theme. Here's a YouTube link for that:
