Description
Before you say, no technically this isn't a redesign, I like the way they look in the show, I just added markings and changed manes mostly with other minor changes, this would be when they're older than after the show ends.

They are in age order! Oldest to youngest. Obviously this is my headcanon, no actual ages have been mentioned in the show.

The mane 6 met when they were older teens, not yet adults. Fluttershy being 19 and Twilight being 17.

General headcanons- Earth ponies are typically more muscular and have a heavier build than other ponies, earth ponies are generally the strongest which helped them with farming when most earth ponies would work on farms. Pegasi are typically slender and light, stereotypically being the smallest of all the pony races, they need to be light in order to fly and so their metabolism is very high, pegasi adapted for speed often have short sleek coats to make them more aerodynamic. Unicorns are more likely to be part of the 'higher class' as there used to be mostly unicorns in royalty and leader positions, they don't usually have much muscle as they weren't used to working physical jobs, and often prefer to use their horns over their hooves. Things have changed since ponies came together and stopped separating from each other. Pure breeds of any of these races are extremely rare and most ponies now have genes from each race.

Fluttershy- The eldest of the group, which is often a shock for lots of people to learn, and a pegasus. She is the 3rd tallest of the 6. Not much change to her in general. She has always been pretty slender, she has long legs and so it makes her slightly taller than the average pegasus mare. Both her mother and father are pagasi and she also has a younger pegasus brother names Zephyr Breeze. She is likely to be descended from a unicorn, perhaps a grandparent. Fluttershy has never been much of a strong flier, but living in the clouds meant she had to learn. She was placed into a young fliers camp, meeting a younger filly names Rainbow Dash who took Fluttershy under her wing and helped her out when team leaders neglected her and classmates teased her. She found it difficult to make friends and stuck to Dash's side, even though her friend was the best flier in the class and she was the worst, Dash would always help her with basic flying and Fluttershy would support Dash's tricks and stunts (being the one to go running to an adult when Dash inevitably hurt herself). Being knocked off a cloud during a race she was watching made her come to realise her talent, animals. She wanted to improve her flight skills even more so she could visit her animal friends whenever she wanted, eventually dropping out of flight school when she knew she could make it that far. She moved to ponyville as soon as possible, buying a small cottage on the outskirts of Ponyville where she could keep and care for animals.

AppleJack- Second oldest of the group. She is extremely tall and muscular, towering over most ponies. Tallest of the 6, only beating out Twilight by a couple centimetres after the unicorn becomes a princess. She is a purebred Earth pony, her family comes from a long line of Earth ponies, the apple family being one of the most famous purebred families around. Her entire family are Earth ponies, her mother and father both passed away when she was very young, leaving her to live with her paternal grandmother and her older brother, Big Mac, and younger sister, Apple Bloom. As a foal AppleJack was close to her father and was always desperate to work with him, even when she was nowhere near strong enough to properly help, her father would still find little jobs for her to do to feel helpful. When her parents passed when she was around 6 years old, she couldn't properly process that her parents weren't coming back. She and Big Mac were very close in age and had each other for support. Shortly after their passing she was unsure of her place on the farm and went to live with her mother's sister and her husband, Aunt and Uncle Orange in the city, thinking she may figure out who and what she was supposed to do. She didn't spend long with them before realising she belonged on the farm and never had any doubts that it was where she was supposed to be. She became a huge inspiration to her little sister, Apple Bloom who was born shortly before their parents passed and unfortunately has no memory of them. She became a slight maternal figure to her sister, as Granny Smith was already very old by the time Apple Bloom was growing up, and AJ would be the one to play and take her sister everywhere. AppleJack has a VERY strong family connection, to her, family is everything, so she never likes to be far from home for long periods of time.

Rarity Belle- The third oldest of the group. She is around average height for a unicorn and very beautiful indeed. She is a purebred unicorn born and raised in Ponyville but wishes to become upper class or even royalty someday. Rarity was raised by both her unicorn parents, she got her cutie mark after the sonic rainboom caused her to find gems inside a rock. She used these gems to create clothing for ponies, which ponies bought from her as a filly, and she loved it. Her little sister Sweetie Belle was born when she was around 8. Once Rarity was a little older, her parents would start leaving her to look after Sweetie Belle whilst they went out. They were travellers and didn't want to put their holidays on hold for until Sweetie was old enough to be alone herself. Rarity spent most of her early teen years caring for Sweetie and the two are very close, even though they fight quite often when Sweetie Belle is making Rarity lose focus from her work. Sweetie Belle looks up to Rarity more than her own parents. Rarity realised a little later on that what her parents did wasn't normal and she lot out on a lot of her social life as a kid because of it. She is a famous dress maker, opening different boutiques around Equestria, obviously not running or being the main dress maker herself for all of them (seriously, what a ridiculous standard lol) She travels to check up on each store now and again, and is still the main designer of the company.

Rainbow Dash- Third youngest of the group, only a month or two older than Pinkie. She was named after her rainbow mane and need for speed ever since she was a foal. She was sent to a flight school as soon as she was old enough, and soon surpassed even the older pegasi in skill and speed. She was always training, it was all she ever wanted to do and she seemed to have endless energy when it came to flying. Despite being the best flier in school, she made an instant connection with an older classmate, Fluttershy, and would help support her and teach her the basics of flight, she could tell Fluttershy wasn't being taught well in class and wanted to help her friend. In a race as a young foal, she made history with a 'sonic rainboom' and was an extremely popular kid, forever known for the stunt she pulled as a filly. Fluttershy stopped showing up to flight school eventually, but the two still remained friends. Dash bought her own cloud home so she was able to move wherever she wished, cloudsdale was always her home and she never wanted to live on the ground as a kid. She trained whenever she could to become part of her favourite areal stunt group, the Wonderbolts. As soon as she was old enough, she applied and entered the trainee stage. Eventually passing to become a fully fledged Wonderbolt after a member retired and a position opened up. She considers herself 'living the dream' as she is in her childhood dream job with amazing friends and support. She is an only child, both parents are pegasi. Her mother Windy Whistles is likely a pure bred pegasus, her short build was inherited by Dash. Her father, Bow, is the son of a purebred pegasus and an earth pony, his muscular build giving him some flight difficulties, meaning he isn't as agile as other pegasi and prefers walking rather than flight.

Pinkie Pie- The second youngest of the mane 6, though only a year younger than Fluttershy, she is older than Twilight by around half a year. She is a very very small pony, especially for an earth pony. She has pegasi genes from her granny pie (maternal), which is where her pink colouration comes from. Her family are rock farmers who live in a very isolated area not many ponies know of. She always felt pretty out of place in her family, her bright coat and mane made her stand out, and she always knew she acted nothing like her stoic, serious family members. She left home when she was around 12, after pleading with her parents, who agreed she wouldn't be much use on the farm as she grew, her cutie mark being nothing to do with rock farming as well as her small size. She made it to Ponyville where she found Mr and Mrs Cake, Cupcake being her maternal aunt. She was happily taken in by her Aunt and Uncle, and threw parties for everypony, instantly knowing what she was meant to do. The size of Ponyville was small so she always knew every pony around and her happiness only grew the longer she stayed. Her father's family owns the rock farm and he had an arranged marriage with her mother, knowing they were to be wed when they were still young. Their marriage is nowhere near perfect as Cloudy never wanted to work on farm of rocks, but love each other genuinely deep down. Pinkie Pie has 3 sisters, LimeStone, Maud being older and Marble is her twin sister, she always had a strong bond with them and never lost contact, even after she moved away.

Twilight Sparkle- The youngest of the mane 6, moving to Ponyville at 17, and studying under Princess Celestia since she was 4 years old. She hatched her dragon, Spike, in her entrance exam, adopting him as her little brother, a very unexpected decision made by her and her parents. Her father is known as a unicorn bat pony, a bat pony with no wings, but sharp teeth, fluffy ears and slit pupils, he shared his genes with his biological children, Twilight and Shining Armour. Twilight's wings were a magical addition, blessed to her by her mentor, Princess Celestia, the bat wings surprising them both when becoming a princess. Her family were upper middle class, known throughout Canterlot where they resided when Twilight was a foal. When Shining Armour married a Princess, the family technically became extended royalty, though their parents were never interested in status and prefer to think of themselves like every other pony, just having fun without worrying about what other ponies think. Twilight's mother, Twilight Velvet, came from an upper class, pure unicorn family, but she never felt like she fit in. Night Light is a bit of a mixed bag, mostly a unicorn family from one side, and a bat pony/ pegasi/ unicorn family on the other.

WOO, okay I hope all this made sense, it took so long to write up! I really hope you like these gals, I wanted to make this a general headcanon and not for any specific next gen universe (if I end up with more than one lol) so all of this is just stuff that happens before the show pretty much.

Some random notes:
- Scootaloo isn't related to Dash in any way, they aren't relatives, Scoots just look up to Dash a lot, being her idol, she just wishes she was her sister. This is why she isn't mentioned in Dash's section lol. Dash is very fond of her, but no, she doesn't adopt her. - The age gap between Fluttershy and Twilight is 2 years, so all of their ages are within 2 years of each other lol.
- The CMC bond over having absent parents- Apple Bloom's being deceased, Sweetie Belle's being absent and Scootaloo with one deceased and one absent parent. Rarity and AJ think it's nice they can all comfort each other with their similar situations.
- Fluttershy actually met AJ through her animals. They became close friends, and Fluttershy is always the first pony AJ goes to if she is worried about an animal.
- Pinkie, AJ and Rarity all met at the Ponyville school. Rarity and AJ meeting as soon as school started, and Pinkie meeting them when she moved into Ponyville.
- Rarity's parents don't hate their daughters by any means, but both of them were accidental pregnancies, and the two weren't really meant for parenthood. This doesn't excuse their behaviour however. They are travellers, not wanting to spend lots of time in one place, and so raising their two daughters for just over a decade was way too long for them, and they instantly got back into travelling. (Yes, they left Rarity to look after Sweetie Belle WAY earlier than they should have, without any other supervision being arranged. Luckily they live in a small village so a lot of ponies were willing to help them out. But they were VERY wrong to do this.)
- Rarity and AJ actually didn't get along at first, Rarity used to think AJ was messy and didn't have proper manners, and AJ thought Rarity was rude and stuck up. They actually got into a lot of arguments. It wasn't until they were forced to hang out together for a project outside of school (the teacher TOTALLY didn't do that on purpose) that they started to get along. They still butt heads due to their different outlooks, but they're good friends.
- Dash's parents actually struggled to conceive. They both wanted a lot of kids, having so much love to give, but after many failed attempts, once they had Dash they realised how much a miracle she was and decided to put all their love and attention on her, spoiling her to no end. They considered maybe trying one more time, but Dash was indeed a huge handful and they didn't want to go through the intense emotions of failing to conceive another child again. This is why they're very intense to say the least, when it comes to their daughter, and Dash has never once doubted her parent's love.
- AJ probably would've had more sibling if her parents hadn't passed away. They passed from a timberwolf attack one night, after trying to protect the farm. The whole situation is very traumatising to the family and they only speak of their parents fond memory, not how they passed.
