I was actually thinking of a death battle with characters from Buffy and Rosario+Vampire mixed in.
Spike has hundreds of years of experience while Moka is just a teenager. However, Moka has defeated enemies of immense strength like the Public Safety Commission. Spike has killed two slayers which is a major claim for a vampire. Its hard to determine which accomplishment is better but Spike did it himself while Moka had friends always helping her. Considering this, I say Spike would prove to be the victor. He could probably use some of his tricks to exploit Moka's aggressive nature and lure her into a trap, thus vanquishing her.
