Multiply.Never ever sketch over  
This is part of the noobs mistakes everybody did ,and so i ,lol (back in time of course,which means uh...years ago ) Unless you're trying to turn your sketch in a full digital painting of course
I always use at least 3-4 layers for characters (line + hairs,skin and clothes colors). I used two colors layers on my last pic and that's probably why i'm not satisfied :/

Ahh so you use the multiply ^_^ When I use multiply there always seems to be some extra lines i dont like and the colour doesnt turn out the way I want it. To dark. I like bright, clean colours. 
Huh okay, lots of separate layers for you I want to use as little as possible.

Extra lines ? what do you mean ? Do you clean your sketches before colors ?
Too dark ? A tip if you want to colors inked work ,make sure your black pixels are black,and your white pixels are white. Get rid of the grey ones.Even if you have to scan in Black&white with 600 dpi,then re-adjust it to 400.
If you want to color a traditional pencil picture,i recommend to adjust contrast/brightness so you don't have the grey/badly scanned syndrome and no prob with the colors Do you use curves tool (ctrl+M) ?it's useful : )
Haha,3 layers is nothing,i remember using 30-40,even more (for some pictures) xD  Poor computer

Cha. I get this very grainy look at some extra lines i dont want (like sketchy lines). 
Yeah I use curve tool and level tool. I like levels more but I should try curves then
Wow 30 to 40 XD I used that many too! It slowed my computer down and I hate it XD I like using very little layers and i can apply brush strokes quickly. 
Hope your computer does not suffer from many layers now XP

Why you no get rid of the extra lines with a bit of Dodge/highlight tool ? 
My computer is suffering from a lot of things ,lol It's getting old ,now
