Knuckles is from the Sonic the Hedgehog series.
Donkey Kong is from the Donkey Kong/Mario series.

Interesting note: Both of these characters have appeared together in 3 games. Super Smash Bros. Brawl (were Donkey Kong was a playable character and Knuckles made a cameo appearance as a trophies, sticker and as a background character in the Green Hill Zone stage), Mario and Sonic at the Olympic Winter Games, and Mario and Sonic at the London 2012 Olympic Games (were both characters are playable). They will be appearing together again in the upcoming 4th game of the Mario and Sonic at the Olympics series, Mario & Sonic at the Sochi 2014 Olympic Winter Games.
