Skeletron is the only boss in Terraria with 6000 hp.
He also has two hands that are 1500 hp each, so taking out both arms makes the boss far easier than one would think (one of the arms was actually destroyed by a star falling out of the sky, luck or coincidence, I don't care which).
To face the boss, you must venture to the entrance of the dungeons and speak to the old man at night (7:00PM-4:00AM). Right click him at night and you will enter a battle against him.

The best strategy is elevation. Before facing him, cut down some trees and make wood platforms so you can jump about while you are in combat (I used jet boots obtained earlier in fight with the Goblin army). Jet boots can give a HUGE advantage against this enemy, so if you found some, get 30-60 large health potions, your strongest weapons and armor, and head out for the dungeons.

Defeating Skeletron will give you unlimited access to the dungeons, where you can get the muramasa, blue moon, and cobalt shield, just to name some.
