I dont know what is a "marble" and I havenΒ΄t seen the serie (Only the episode where is based this comic), but I use that "sparkle" as a visual narrative resource.

Ah. marbles are small round things usually made of glass usually coloured or with a twirl or spiral pattern in the middle.
And when someone is looking their marbles, ie. marble falling out of Pinkie Pie's ear/head, it means they are going crazy or losing touch with reality.
