You can go either way. For more painterly techniques, you can still emphasize lineart, but you would want to do it without any sort of line tool. Think of it as sketching. You still darken in portions of your contour, but it's not blocky. That's a bad explanation, I know. To see what I'm talking about, check out this guy's work: [link]
Really, as you get used to adding more contrast and volume to your painting, the painting of your lines will become more instinctive, as well.
Honestly, I'm not a fan of line tools. They're clean, and that's nice, but they strip any sort of realistic weight from your lines, leaving your image somewhat flat no matter how well you shade it.
