I don't think there's a tutorial, hit Ctrl D a few times and it should expand into something. Try clicking the buttons to see what they do. For my stuff, I just use it to get the lines, then save as postscript and open it in Adobe Illustrator and go from there. You might find some instructions on it at the site where you downloaded it too.

Oh! Well, I get the lines. I just didn't know what to do from there. So what do you do in Illustrator with it? I guess I need to start playing around more.

Sometimes I use LivePaint to color it in Illustrator (gotta have CS2 for that, though) then, I take it from Illustrator into Photoshop and play around with it. You can color it in Photoshop as well. The Penrose Tiler software is really just a starting point. Hope that helps.
