And in answer to your question, I save the image in photoshop as a transparent file (.png usually). Hope that helps!

Well, as long as it's transparent. Make sure to not merge down all the layers, just select merge visible and then go to file > save for web. (This is on Photoshop, by the way).

Just don't merge down the layers and it should be transparent. At the beginning, when you create a new document in Photoshop, choose a transparent background.

I don't think having a lock is a problem (in terms of the transparent thing). Just means you can't alter the layer.
When you click on File > New, look at the background contents. There's settings for white, background color and transparent. Select transparent!
