While the X-Patrol (Doom Patrol + X-Men) is the frontline for Meta-Mutants (Meta-Humans + Mutants) and human coexistence, the Brotherhood (The Brotherhood of Evil + The Brotherhood of Mutants) stands for Meta-Mutant supremacy. Many of its members have seen the worse humanity has to offer, thus believe that the extinction of the human race would be best for the Meta-Mutant race.

Although there are a multitude of members, there are four key leaders who organize the group and its many factions. These four include:

Magnetron (Brain + Magneto): Never underestimate willpower and raw, unadulterated rage. Ernst Eisenhardt suffered at the hands of hatred all of his life. Whether it was because he was Jewish, gay, or a Meta-Mutant, he found himself a victim of bigotry for far too long. In his earlier years, he had met a young Chief X (Chief + Professor X), who wanted to create a safehaven for Meta-Mutants. At first, Ernst was more than happy to join Chief X in this plan, until he realized how fruitless it was. Chief X wanted a world where humans and Meta-Mutants could live together in harmony. Ernst, being a victim to hateful humans all his life, rejected this notion, leading to a battle between the two former friends. With Chief X's telepathy and telekinesis and Magnetron's magnetism, the two were locked into a battle of equal strength. The conflict resulted in Chief X becoming crippled, and Magnetron sustaining serious damage to his organs. With his body betraying him, Ernst looked to methods to preserve his life by any means necessary. Conjuring all the strength in his body, he forced the iron in his blood to keep him alive just a little longer, until he was met by the Immortal Mastermind. Having survived for centuries, the Immortal Mastermind had found a multitude of ways to preserve a body. Magnetron's brain was put in a glass jar, but in a surprise to himself and the Immortal Mastermind, he managed to retain his magnetic abilities. Ernst created a new body for himself, made completely of metal, and recruited the Immortal Mastermind to his cause of Meta-Mutant supremacy.

Madame Mystique (Madame Rouge + Mystique): Raven de Mille was born with the incredible Meta-Mutant ability to shapeshift and alter the size of her body. Unfortunately, her shapeshifting would have a horrible drawback. Any time she would transform into a person, she would also take on their personality. This led to her adopting Disassociate Identity Disorder, with the number of her alters growing with each new transformation. Eventually, she was approached by Magnetron and Dr. Dino, who managed to create a serum that could suppress her alters. As a show of gratitude, Madame Mystique agreed to join their team. Of course, each time Madame Mystique would transform into a new person, she would regularly have to take the same serum, making her pool of transformations limited depending on her medical supplies.

Dr. Dino (Monsieur Mallah + Sauron): When scientists discovered a way to revive dinosaurs, you'd think their research would be done there. With such a remarkable achievement, what more could they want? Like many scientists who have played god, you keep pushing. Creating an army of dinosaur soldiers became the top priority for the group of scientists. To do that, they would need to transform the feral minds of the revived fossils into that of warriors. Through several grueling experiments, they managed to make the dinosaurs "more human" but that came at a cost. One of the dinosaurs, nicknamed "Karl", had surpassed the intelligence of his creators; he would not be turned into a reptile with a gun. One night he managed to escape his cell and sabotage the equipment of the scientists. The next day, a horrible fire broke out in the lab, offering Karl and his reptile brothers the perfect escape. Unfortunately, in a fit of rage, the last human standing shot the remaining dinosaurs before Karl managed to rip the man's head off. Karl looked around, horrified, he was the only one of his kind. He had no idea where to go.

After a few poor attempts to blend in with humanity, he eventually caught the attention of Magnetron, who took pity on him. Magnetron saw the potential of Karl and made him his right hand man. The two initially bonded over their hatred of humanity, but over time a more passionate relationship blossomed between the two.

The Immortal Mastermind (General Immortus + Mastermind): With no definitively known name, the true origin of the Immortal Mastermind is unknown. Some believed that he was the first Meta-Mutant, however that would later be revealed to be Mutawahish (Vandal Savage + Apocalypse). Regardless of his true origin, the Immortal Mastermind is a conqueror who has lived for centuries. Because of this, he easily has the best battle strategy known to humanity. It is believed that his experiences with the occult may be the true source of his immortality. That could also be behind his ability to manipulate the five senses. When the Immortal Mastermind casts an illusion, his victims will experience everything as if it was actually there. Because of his remarkable skills and power, he is one of the deadliest members of the team.
