It's never explicitly stated they're actually blood related. Tbh, they don't even need to be; Some people just like to call each other family because they feel like family. No actual family connections needed.
Also, even if they are blood related, it's obviously very distant and at this point there is no shared DNA anymore.

Eh, well My Little Pony's been around since the 80's. Lauren just rebooted the idea, and helped create the new show. Bonnie Zacherle is technically the creator of MLP, which at the time was only toys.
