Your best starting point is to have a look at ^livius 's Making Emotes: The Tutorial Master List . There are tutorials there that cover basic emotes, animation and advanced shading and effects. These tutorials also cover different applications like Paint, Photoshop, Fireworks, PaintShopPro and Gimp.

I use PaintShopPro to create the static parts then animate them in Ulead Gif Animator.
