What does "manifest" mean? Are all the cars going to one place?

They head to a classification/"Hump" Yard and then get routed to different destinations...In Chicago BNSF closed their hump yard and run through to either another railroad or the Belt Railway sorts their cars for further distribution. In Chicago...Proviso (UP) and Bensenville (CP) still hump cars.

A manifest is like a regular freight train with general mixtures of cargo...as opposed to a unit coal train or grain train or intermodal (COFC/TOFC) train
