i use photoshop cs5, but any version of photoshop would be good, or any program that can hold transparency and has a binary/pencil tool. GIMP could do pixels, as well. What I do is sketch out the character on a 100x100 file, it's much easier than sketching on a 50x50 canvas. After finished, I copy the sketch and paste it down onto the 50x50 file. For the lines, I will give you a tutorial since it's hard to explain.
