Well, there's certainly not a lot new going on here. It certainly doesn't make it bad; it just doesn't leave much in the way of surprises.
Now what WOULD have been surprising is if Twilight is about to enter the barn, and Fluttershy stopped her, hearing the noises coming from within. Twilight turns to her with a smirk. "Really, Fluttershy? The old, 'mistake-friends-for-making-sex-noises-when-they're-doing-something-totally-different' gag? Not very original..." She proceeds to open the door, and we see Rainbow mounting Applejack (nothing explicit necessary, but Rainbow hunched over AJ's back from behind would suffice for the impact to be felt). Twilight freezes. So do they. Fluttershy blushes. Twilight asks something like, "Is this not what it looks like?" followed by Rainbow saying, "It kind of is!"
Maybe that wouldn't have been as funny, but it certainly would have kept people guessing.
That being said, the art style is phenomenal as usual, the expressions are nailed perfectly, the backgrounds are suitable, the dialogue is spot-on and worth a smile. As ever, I love some AppleDash, and even if it isn't there, I like it just the same when it's implied.
Not everything has to be original. People who enjoy your art style will like this piece (I do), and people who don't grow tired of rehearing the same joke repeatedly won't mind it either. I hope you consider rehashing it a different way someday, though. I love your AppleDash strips, and I'd like to see your interpretation on their relationship go even further.

I have to add my opinion in along side Mad42Hatter in saying that, honestly, this wasn't very funny or original. While the joke is indeed classic, it has been done to death in a thousand different ways to the point where it's just beating the ground where the dead horse decomposed at this point.
Now, if it were the anyone else I might let it slide, but you're better than this, Red. You've come a long way in your comics since you started, to the point where I believe I can expect better from you and most the time you give it. I don't want to seem overly harsh, but I just think you could do much better.
That said, I do give points for improving some of your art. I think the facial expressions are even better than usual, and I like your use of backgrounds both in color and style that complement the piece. I also think your placement of speech bubbles has improved to the point where you seem to place them strategically rather than willy-nilly, so points for that. Now what would have been more original... is if you'd taken Twilight as per the usual, and Fluttershy tried to stop her. Twilight goes, "Really, Fluttershy? Twilight kicks the door open, and we get Rainbow in a mounting-position over Applejack. (Let's not make it explicit; some things are better left to the imagination.)
Twilight goes, "Uhhh... is this not what it looks like?" Rainbow says, "It kind of is?" And... something else humorous. I dunno. I'm tired.

I mean, can't we all agree that at this point in human history we've pretty much done it all and we're just tweaking things and slightly repackaging things? And, you know, sure.Every once in a while there's some genius who comes along and reinvents the way we all look at things, but that's not most of us. We can't all aspire to that.
Yes it's unoriginal. Your describing it as such means nothing. Doesn't matter.
Is it good? Does it entertain you? Does it make you feel something?
Why do you care if something slightly similar has been done before if it's effective?

Well, deviantart makes people fill out Originality ratings up there for a reason. What, do you think everybody's just supposed to automatically get 5 stars?

Don't tell me to calm down. If he's allowed to have a stupid opinion then im allowed to call him on it. If he didn't want people to debate his rating then he shouldn't have made an official rating. Expecting nopony to think otherwise than you just because you are reviewing something doesn't make you exempt from criticism.

Red can do much better than this. Yes, originality is hard to come by these days, but if your work starts turning into just another rehash of a joke that's been done 100,000 times in this fandom alone, your quality will decay.
It shouldn't be effective. With a few exceptions (IMO), Red has made some pretty hilarious stuff. Only 12-year-old boys and those who cannot grasp upper-brow humor will should find this even remotely amusing.

No, I don't. But you don't even have to pretend to speak for everyone to know what kind of humor 12-year-old boys like, and what kind of humor is banal and low-brow. They are one and the same.

If they don't want their criticisms to be subjected to scrutiny, then they should't make them as an official review and leave it as a simple comment.

It does not matter if a gag has been done to death or not. Really, it doesn't. For one thing it will always be new to someone.
More importantly though even someone who has heard the joke can find it funny depending on the setup, timing, and delivery. Kind of like how good acting and direction can make a old story feel fresh.
Also, half the fun of this type of gag is the double entendre in the participants dialog. I will grant you however we didn't get any really good lines.
