Yes, he's got heterochromia. The black scleras are natural too, idk, i think it's some kind of mutation or something. (but really, it's only for the aesthetic)

Probably from Joker's mutated DNA. I'm pretty sure that acid did more than just drive him crazy, and make him look like a clown. Some versions of Joker have him, not only immune to any and all poisons and toxins, but also his bodily fluids are toxic as well, such as his blood, when a Mosquito once bit him, and choked to death on the deadly blood it ingested.

One of the coolest things, I think Joker ever did, was during a team up with Scarecrow, Crane double-crossed him, and sprayed him with his Fear Gas, because he wanted to find out what the Joker was afraid of, but to his horror, the gas did absolutely nothing to him, and then Joker gleefully beat Scarecrow up with a chair.
