If my math is right, April should be within her early 20's, since she had been training with Master Splinter and the Turtles for 4 years; starting at 16 years of age with she made her appearance.

At the beginning of the show she was 15, like the turtles, but I do not think that this is really important. My calculations may also be wrong, because it doesn't matter either.

No offense, but my math can be wrong at times as well. I thought it would be worth pointing out that when she was gifted her Tanto by Splinter, she had been training for 4 years. And she had been given the weapon in season 4, which would account for the 4 year timeline. So if memory serves me right, the 5th, and final season she would be probably be in her early 20's. Give or take a year.
Would anyone like to put in their thoughts in that?
