A Nuzlocke is a run through any Pokemon game with these two added rules:
1. You can only catch the first Pokemon found in a route/area.
2. If a Pokemon on your team faints, it is considered dead and cannot be used for the rest of the game.
It sounds kind of morbid, but it is actually an extremely addictive added challenge. I have about eight ongoing Nuzlockes right now.
Many people add on different rules, like no catching Pokemon they already have, or rules addressing which items can be used, just to add more challenge or to give their Nuzlocke a bit of a twist.
If you lose your entire team or all Pokemon in your box, the Nuzlocke is considered a loss. If you defeat the Elite Four, you have completed the Nuzlocke.

Ah, but Nuzlocking makes a person much more careful, even in normal Pokemon runs.
You'd be surprised how far you can go with some caution (and Potions. XD )
