Yes - she was an alicorn in Twilight Sparkle's flashback in 'A Canterlot Wedding', well before Twilight got her cutie mark. (Though this raises a question of why did a princess foalsit a seemingly ordinary unicorn. This is [perhaps intentionally] only vaguely explained - Cadance herself predicted that Twilight was "anything but a regular old unicorn".)
